Overview

Methyl-CpG binding domain protein 2 (MBD2) mRNA is the messenger RNA transcript that encodes the MBD2 protein, a key reader of DNA methylation. The MBD2 protein specifically binds to methylated CpG dinucleotides and recruits the NuRD (Nucleosome Remodeling and Deacetylase) corepressor complex, leading to chromatin condensation and transcriptional silencing of target genes (UniProt P35209). In several diseases, including various cancers and systemic lupus erythematosus, MBD2 is overexpressed, which leads to the aberrant silencing of tumor suppressor genes or the dysregulation of immune-related genes (PubMed: 11544176, 18205210). Therapeutic strategies targeting MBD2 mRNA, such as antisense oligonucleotides like MG98, aim to reduce the levels of MBD2 protein to reverse these epigenetic changes and restore normal cellular function (PubMed: 15507677). While clinical trials have explored these RNA-targeted therapies in oncology, achieving high specificity and efficient delivery remains a primary challenge in the development of MBD2-targeted agents (PubMed: 12473715).
