Target intelligence / Profile preview
The MHC class II-restricted T cell receptor (TCR) recognizing CRM197-derived peptides is a specialized immune receptor found on CD4+ T lymphocytes. CRM197 is a non-toxic mutant of the diphtheria toxin widely utilized as a carrier protein in conjugate vaccines to enhance immunogenicity against bacterial polysaccharides (Source: PubMed PMID 27511734). These TCRs play a pivotal role in the "carrier effect," where they recognize CRM197 peptides presented by Major Histocompatibility Complex (MHC) class II molecules on antigen-presenting cells (Source: UniProt P00588). This recognition event triggers T cell activation, providing the necessary signals, such as cytokines and CD40L expression, to B cells for high-affinity antibody production and immunological memory (Source: StatPearls, Vaccine Adjuvants and Carriers). In clinical practice, these receptors are the functional targets of widely used vaccines like Prevnar 13 and Menveo, which rely on the pre-existing or induced T-cell memory against CRM197 (Source: FDA, Prevnar 13 Package Insert). The interaction between the TCR and the CRM197-peptide-MHC complex is a critical determinant of vaccine efficacy across different populations.
The TCR recognizes CRM197-derived peptides presented by MHC class II molecules on antigen-presenting cells, leading to CD4+ T cell activation, cytokine secretion, and the provision of help to B cells for antibody production.
