MicroRNA 140-3p (miR-140-3p) is a small, non-coding RNA molecule that plays a critical role in the post-transcriptional regulation of gene expression (PMID: 20018668). It is highly enriched in chondrocytes and is essential for cartilage homeostasis and skeletal development by suppressing proteases such as ADAMTS5 and MMP13 (PMID: 25100639). In addition to its chondroprotective role, miR-140-3p functions as a tumor suppressor in several cancers, including lung, breast, and gastric cancer, where it inhibits cell proliferation and epithelial-mesenchymal transition (PMID: 28656235). In osteoarthritis, the expression of miR-140-3p is significantly reduced, contributing to the progression of joint degeneration (PMID: 24557190). Therapeutic approaches currently under investigation include the use of miR-140 mimics to restore its levels in diseased tissues or antagomirs for specific regulatory interventions (PMID: 30210140). Despite its potential, therapeutic application faces challenges such as ensuring stable delivery to target tissues and minimizing off-target effects (PMID: 23913143).
