Overview

MicroRNA 828b (miR-828b) is a small non-coding RNA molecule primarily recognized for its regulatory role in plants, where it controls anthocyanin biosynthesis and trichome development by targeting MYB transcription factors [1, 12, 14]. In the context of human therapeutics and cosmetics, miR-828b has emerged as a bioactive component of tea leaf-derived nanovesicles (TLNVs) capable of cross-kingdom regulation [6, 8]. When applied topically to human or mouse skin cells, miR-828b directly targets the MYB4 axis, which modulates the PI3K/AKT signaling pathway to downregulate key melanogenesis regulators such as Microphthalmia-associated transcription factor (MITF) and tyrosinase (TYR) [6, 8]. This interaction effectively reduces melanin production and epidermal thickness, offering a potential therapeutic strategy for hyperpigmentation disorders and UV-induced skin damage [8, 10]. Beyond pigmentation control, miR-828b delivered via plant-derived vesicles exhibits antioxidant and anti-inflammatory properties by suppressing inflammatory cytokines like TNF-α and IL-1β and promoting melanoautophagy [8, 10]. While traditionally considered a plant regulator, its application in human nanotherapeutics represents a novel approach to utilizing natural RNA molecules for precision dermatological treatment [6, 8].

Mechanism of action

miR-828b mediates post-transcriptional gene silencing by binding to the 3'-UTR of target mRNA (such as MYB4 in mammalian cells), leading to mRNA degradation or translational inhibition, which subsequently suppresses the PI3K/AKT signaling pathway and reduces the expression of melanogenic factors like MITF and tyrosinase.
