MicroRNA let-7c (miR-let-7c) is a highly conserved member of the let-7 family that serves as a master regulator of gene expression by binding to complementary response elements (MREs) in the 3' untranslated regions of target mRNAs [1, 2]. These response elements are critical regulatory nodes in oncogenic and pro-inflammatory transcripts, including KRAS, MYC, and IL-6 [3, 4]. By targeting these elements, miR-let-7c induces translational repression or mRNA degradation, thereby acting as a potent tumor suppressor and anti-inflammatory agent [5, 6]. In many pathological states, such as lung cancer or chronic inflammation, miR-let-7c is downregulated, allowing for the uncontrolled expression of its targets [7]. Therapeutic strategies involve the use of miR-let-7c mimics to restore this regulatory control and suppress the production of oncogenic and pro-inflammatory proteins [8]. However, the clinical application of these mimics faces challenges such as efficient delivery to target tissues and the potential for off-target effects or innate immune activation [9].
