Overview

The Microtubule affinity-regulating kinase 1 (MARK1) mRNA 3' untranslated region (3'UTR) is a non-coding sequence at the end of the MARK1 transcript that regulates protein synthesis (UniProt Q9P0L2). It serves as a scaffold for microRNAs (miRNAs) and RNA-binding proteins that dictate the half-life and translational efficiency of the mRNA. The MARK1 protein itself is a serine/threonine kinase that plays a vital role in microtubule dynamics by phosphorylating microtubule-associated proteins (MAPs). Research has highlighted the 3'UTR as a susceptibility locus for autism spectrum disorder (ASD), where specific SNPs like rs2233152 are associated with altered expression levels (Maussion et al., 2008; PubMed: 18332877). In the context of cancer, the 3'UTR is targeted by miRNAs such as miR-155, which can lead to the downregulation of MARK1 and subsequent changes in cell polarity and migration. Because MARK1 levels are critical for proper neuronal migration and dendrite development, the 3'UTR is a key site for post-transcriptional control. Therapeutic strategies targeting this region include the use of antisense oligonucleotides (ASOs) to modulate protein expression. Experimental microRNA mimics or antagomirs are also being explored to correct dysregulated MARK1 expression in disease states. This regulatory region is thus a promising target for precision medicine in neurodevelopmental and oncological disorders.
