The colchicine-binding site is a specific region on tubulin, the building block of microtubules. Binding of colchicine or other inhibitors to this site prevents proper polymerization of tubulin dimers into microtubules, disrupting normal microtubule dynamics and inhibiting cell division. This mechanism is exploited in cancer chemotherapy.
