Overview

Microtubule-associated protein 2 (MAP2) is a cytoskeletal protein primarily localized in the dendrites and cell bodies of neurons, where it plays a critical role in stabilizing microtubules and maintaining neuronal morphology (Sanchez et al., 2000). The term 'Microtubule-associated protein positive nerve fibers' refers to a histological classification used in immunohistochemistry to identify and quantify nerve density, particularly in studies of neuroregeneration or neurotoxicity (Johnson & Jope, 1992). While MAP2 is essential for the stabilization of the neuronal cytoskeleton and facilitates intracellular transport, it is not a direct therapeutic target for current drug interventions. Instead, the density and integrity of MAP2-positive fibers serve as important biomarkers for assessing neuronal health and the efficacy of neurotrophic factors in diseases such as Alzheimer's, stroke, and traumatic brain injury (Soltani et al., 2005). Changes in MAP2 expression levels often correlate with synaptic loss and cognitive decline, making it a vital readout for neuroplasticity (Dehmelt & Halpain, 2005). Although related proteins like Tau (MAPT) are therapeutic targets in tauopathies, MAP2 is predominantly utilized as a structural marker in preclinical and clinical pathology research.
