Overview

The MICOS-MIB-ANK2 complex is a specialized protein assembly that bridges the inner and outer mitochondrial membranes to maintain the structural integrity of mitochondrial cristae. The Mitochondrial contact site and cristae organizing system (MICOS) and the Mitochondrial Intermembrane space Bridging (MIB) complex are essential for cristae junction formation and protein import (Yang et al., 2023, Nature). Recent research has identified Ankyrin-2 (ANK2, also known as Ankyrin-B) as a critical scaffolding component that organizes this complex, facilitating the stabilization of MICOS-MIB and regulating cardiolipin homeostasis (PMID: 37495689). This interaction is particularly vital in high-energy tissues like the heart, where it ensures efficient ATP production and metabolic stability. Mutations in ANK2 or disruptions in the MICOS-MIB assembly are linked to severe pathologies, including arrhythmogenic cardiomyopathy and mitochondrial myopathies. While no drugs currently target this complex directly, compounds like Elamipretide, which stabilize cardiolipin and cristae structure, may indirectly modulate its function. This complex represents a novel therapeutic avenue for treating cardiac and metabolic disorders associated with mitochondrial structural defects.
