Overview

MT-ND4 pseudogene 23 (MTND4P23) is a nuclear-encoded nonfunctional DNA sequence derived from the mitochondrial ND4 gene. Unlike the functional MT-ND4 gene, which encodes an essential subunit of the mitochondrial Complex I involved in oxidative phosphorylation, MTND4P23 does not produce a protein or participate in electron transport or cellular energy metabolism. Pseudogenes like MTND4P23 may be detected in genomic studies or PCR reactions but have no known biological function, therapeutic relevance, or disease association. Their presence reflects historical gene transfer events from mitochondrial to nuclear DNA but they are not considered therapeutic targets, receptors, or biomarkers. MT-ND4 pseudogenes such as MTND4P23 can confound experiments aimed at detecting mitochondrial sequences in nuclear DNA, especially in cell models lacking mitochondrial DNA (rho° cells). However, they do not provide functional compensation or therapeutic utility. Mutations in the actual MT-ND4 gene cause mitochondrial diseases, but pseudogenes like MTND4P23 do not contribute to these phenotypes.
