Mitogen-activated protein kinase kinase kinase kinase 4 (MAP4K4) mRNA is the transcript encoding a serine/threonine kinase that acts as a key mediator in the c-Jun N-terminal kinase (JNK) signaling pathway [UniProt, O95819]. MAP4K4 is involved in various cellular processes, including cell migration, cytoskeletal reorganization, and the inflammatory response, particularly in response to tumor necrosis factor-alpha (TNF-α) [PubMed, PMID: 17446437]. In the context of metabolic disease, MAP4K4 mRNA expression is linked to insulin resistance, as its protein product inhibits insulin signaling in adipose and muscle tissues [NCBI Gene, 9448]. In oncology, elevated MAP4K4 levels promote tumor cell invasion and metastasis across multiple cancer types [PubMed, PMID: 24510477]. Targeting the mRNA via RNA interference (siRNA) or antisense oligonucleotides (ASOs) aims to silence the gene expression, thereby reducing the pathological effects of the MAP4K4 protein [PubMed, PMID: 19244111]. This approach is being explored for treating type 2 diabetes, inflammatory disorders, and certain cancers where small molecule inhibition of the kinase domain may lack sufficient specificity [PubMed, PMID: 29109508]. Research has also identified MAP4K4 as a potential target in neurodegenerative diseases like ALS, where mRNA knockdown protects motor neurons [PubMed, PMID: 24931610].
