Overview

MLLT1 encodes the protein ENL, a chromatin reader component of the super elongation complex (SEC) that increases the catalytic rate of RNA polymerase II transcription by suppressing pausing and binding acetylated or crotonylated histones via its YEATS domain, with preference for H3K27cr. It acts upstream of negative regulation of protein kinase activity, is located in nucleoplasm and cytosol, and is implicated in oncogenic gene transcription in acute myeloid leukemia. Gene fusions, mutations, and rearrangements are observed in various cancers including myeloid/lymphoid leukemia, eye cancer, intestinal cancer, and skin cancer.
