Mohawk homeobox (MKX) is a member of the Three Amino acid Loop Extension (TALE) family of homeobox transcription factors, primarily recognized for its pivotal role in the development and homeostasis of tendons and ligaments (UniProt Q86WJ1). It is expressed in the progenitor cells of the musculoskeletal system, where it regulates the transcription of essential extracellular matrix components, including type I collagen and tenomodulin (Ito et al., 2010, Nature Communications). Research indicates that MKX is necessary for the proper alignment and maturation of collagen fibers, and its deficiency leads to hypoplastic tendons and increased susceptibility to injury (Liu et al., 2015, PNAS). In the context of disease, MKX downregulation is linked to tendinopathy and intervertebral disc degeneration, making it a significant target for regenerative medicine (Nakamichi et al., 2016, Nature Communications). Current therapeutic explorations involve using MKX-based gene therapy or mRNA delivery to stimulate tissue repair and restore mechanical properties in damaged musculoskeletal tissues (Nakamichi et al., 2018, Science Signaling).
MKX acts as a transcriptional activator that binds to specific DNA sequences (e.g., TGACAG) to promote the expression of tenogenic genes such as Type I Collagen (COL1A1), Scleraxis (SCX), and Tenomodulin (TNMD), thereby driving the differentiation of mesenchymal stem cells into tenocytes and maintaining tendon homeostasis (PubMed: 20461080).
