The Mouse major histocompatibility complex class I H-2Kb presenting Ovalbumin peptide 257-264 (SIINFEKL) is a well-characterized peptide-MHC (pMHC) complex used extensively in immunological research. It consists of the H-2Kb heavy chain, beta-2 microglobulin, and an 8-amino acid fragment of chicken ovalbumin (SIINFEKL) that binds with high affinity to the H-2Kb peptide-binding groove (Rotzschke et al., 1991; PMID: 1651229). This complex is the specific ligand for the OT-I T-cell receptor, making it a cornerstone for studying CD8+ T-cell development, activation, and memory (Hogquist et al., 1994; PMID: 7912655). In the context of drug development, it serves as a target for evaluating novel immunotherapeutic strategies, such as TCR-engineered T cells, cancer vaccines, and TCR-like antibodies (Porgador et al., 1997; PMID: 9115470). By presenting a defined antigen, it allows for the precise measurement of immune responses in preclinical mouse models of cancer and infection. The complex is also utilized to investigate the mechanisms of antigen processing and presentation, as well as the factors influencing T-cell exhaustion in chronic disease states.
The complex presents the SIINFEKL epitope to the OT-I T-cell receptor, initiating a signaling cascade through the CD3 complex that results in T-cell activation, proliferation, and cytotoxic effector function.
