Overview

This target profile represents a group of Type I transmembrane proteins known as Receptor Tyrosine Kinases (RTKs), including the Vascular Endothelial Growth Factor Receptor (VEGFR) family, Platelet-Derived Growth Factor Receptor (PDGFR) family, Fibroblast Growth Factor Receptor 1 (FGFR1), KIT, FLT3, RET, and CSF-1R (UniProt P17948, P16234, P10721). These receptors are critical mediators of signal transduction pathways that regulate cell growth, survival, differentiation, and angiogenesis (Nature Reviews Cancer, 2002). In many malignancies, these RTKs are overexpressed or constitutively activated by mutations, driving tumor progression and the development of a supportive vascular network (PubMed: 17003811). Drugs targeting this specific multi-kinase profile, such as sunitinib and pazopanib, are small-molecule inhibitors that compete with ATP for binding to the intracellular kinase domain (PubChem CID 5329102). By inhibiting multiple pathways simultaneously, these agents can overcome redundant signaling mechanisms and disrupt both the tumor cells and the surrounding microenvironment (StatPearls: Sunitinib). Clinical applications include the treatment of renal cell carcinoma, gastrointestinal stromal tumors, and pancreatic neuroendocrine tumors (FDA Label: Sutent). However, the broad inhibition of these receptors in healthy tissues leads to characteristic toxicities such as hypertension, fatigue, and hand-foot skin reaction (PubMed: 21642684). Monitoring for biomarkers like KIT mutations or RET fusions is often essential for selecting patients most likely to benefit from these therapies (Journal of Clinical Oncology, 2015).
