MYPOP is a Myb-related transcription factor and DNA-binding protein that acts as a transcriptional repressor by binding to the specific core sequence 5'-YAAC[GT]G-3'. It regulates gene expression by silencing viral and cellular promoters, including the long control regions of oncogenic human papillomaviruses (HPV), playing a critical role in intrinsic host antiviral defense and tumor suppression. MYPOP is highly expressed in epithelial tissues and is known to suppress early gene expression and cell proliferation in HPV-infected and transformed cells. Its repressive actions are reduced upon dimerization with PFN1 (profilin), and it can shuttle between the nucleus and cytoplasm. Downregulation or degradation of MYPOP is seen in HPV-transformed cell lines and cervical cancer, supporting a role in oncogenesis when its function is compromised[1][2][3][4][5][6][7]. There are no current drugs, clinical biomarkers, or safety concerns directly associated with MYPOP.
