Overview

BNT164a1 is an investigational mRNA-based vaccine candidate developed by BioNTech for the prevention of tuberculosis (TB) (BioNTech, 2022). It encodes a selection of immunodominant antigens from Mycobacterium tuberculosis, the causative agent of TB, encapsulated within lipid nanoparticles (ClinicalTrials.gov, NCT05537038). Upon administration, the mRNA is taken up by host cells and translated into bacterial proteins, which are then presented to the immune system to elicit a robust and durable immune response (Kaufmann, 2023). This response includes the activation of both CD4+ and CD8+ T cells, as well as the production of specific antibodies, aimed at providing protection against M. tuberculosis infection and progression to active disease (WHO, 2023). The multi-antigen approach of BNT164a1 is designed to enhance the breadth and efficacy of the immune response compared to traditional vaccine platforms like BCG by targeting multiple stages of the bacterial life cycle (BioNTech, 2022). This candidate is currently being evaluated in Phase 1 clinical trials to assess its safety and immunogenicity in healthy adults (ClinicalTrials.gov, NCT05537038).
