Overview

Phytic acid, also known as myo-inositol 1,2,3,4,5,6-hexakisphosphate (IP6), is a naturally occurring polyphosphorylated carbohydrate found in high concentrations in plant seeds and mammalian cells (PubChem CID 890). In humans, it serves as a critical intracellular signaling molecule involved in diverse processes such as mRNA export, DNA repair, and vesicle trafficking (Shears, 2001, Cell). Pharmacologically, phytate is recognized for its potent ability to inhibit the formation and growth of hydroxyapatite crystals, the primary component of pathological calcifications (Schantl et al., 2017, Nature Communications). This property has led to its development as a therapeutic agent, most notably in the form of SNF472, which is being investigated for the treatment of calciphylaxis and vascular calcification in patients with end-stage renal disease (Perelló et al., 2018, Br J Clin Pharmacol). While it offers therapeutic benefits, phytate also acts as an antinutrient by chelating essential minerals like calcium, iron, and zinc, which can lead to deficiencies if dietary intake is high and mineral absorption is compromised (Lopez et al., 2002, Int J Food Sci Technol).

Mechanism of action

Phytate inhibits the formation and growth of hydroxyapatite crystals by binding to their surfaces (Schantl et al., 2017), and it also acts as an intracellular signaling molecule that modulates protein function and gene expression (Shears, 2001).
