N-acetyl-D-glucosamine (GlcNAc) is a monosaccharide derivative of glucose and a key amino sugar found in various biological structures, including bacterial cell walls (peptidoglycan), fungal chitin, and mammalian glycosaminoglycans like hyaluronic acid [7, 15]. In humans, it serves as a vital precursor for the synthesis of proteoglycans and glycoproteins, playing a crucial role in maintaining the structural integrity of connective tissues, joints, and the intestinal mucosal barrier [1, 4]. Beyond its structural functions, GlcNAc is a central substrate for O-GlcNAcylation, a dynamic post-translational modification that regulates protein function, signaling, and gene expression in response to nutrient availability [1, 10]. Therapeutically, GlcNAc is utilized as a dietary supplement for conditions such as osteoarthritis and inflammatory bowel disease (IBD), where it is thought to support tissue repair and modulate inflammatory responses [6, 11]. Emerging research also highlights its potential in treating multiple sclerosis by promoting N-glycan branching on T-cells, thereby dampening autoimmune activity [1, 2]. While generally considered safe, it may interact with anticoagulants like warfarin and is a known allergen for individuals with shellfish sensitivities [6, 9].
