Overview

The N-methyl-D-aspartate receptor (NMDA receptor, or NMDAR) is a type of ionotropic glutamate receptor that plays a central role in excitatory neurotransmission, synaptic plasticity, learning, and memory in the central nervous system. The GluN2B subunit is one of the regulatory subunits that combine with two obligatory GluN1 subunits to form functional NMDA receptors. GluN2B-containing NMDARs are considered promising therapeutic targets due to their involvement in neurodevelopmental disorders, neurodegeneration, chronic pain syndromes, depression/anxiety disorders—and because selective antagonism may offer clinical benefits with fewer side effects compared to non-selective blockade.
