Overview

The glycine site on the NMDA receptor is a binding domain on the GluN1 subunit that is essential for receptor activation. Glycine or D-serine must bind to this site simultaneously with glutamate binding to the GluN2 subunit for the ion channel to open. The NMDA receptor plays a critical role in synaptic plasticity, learning, memory, and neurodevelopment. Dysfunction of the glycine site has been implicated in several neurological disorders.
