Overview

The NMDA receptor is a ligand-gated ion channel in the central nervous system that requires the binding of glutamate and glycine (or d-serine) for activation. The glycine binding site is located on the GluN1 subunit. Activation leads to an influx of Na+, K+, and Ca2+ ions through the channel pore. Fluctuations in extracellular levels of either glutamate or glycine/d-serine can strongly influence NMDA receptor activity, impacting normal brain function and pathological conditions such as neurodegeneration or psychiatric disorders.
