Overview

NAD(P)H pyrophosphatase NUDT13, mitochondrial (NUDT13) is a mitochondrial enzyme belonging to the Nudix hydrolase family[1][3][6][7]. It preferentially hydrolyzes reduced pyridine nucleotides, specifically NADH to NMNH and AMP, and NADPH to NMNH and 2',5'-ADP, with much lower activity towards other nucleotide substrates[1][3][4]. NUDT13 is targeted to mitochondria via an N-terminal mitochondrial targeting sequence, and functions in the regulation of mitochondrial NADH and NADPH levels as well as the NAD+/NADH and NADP+/NADPH ratios, thereby influencing redox state, energy homeostasis, and potentially responses to oxidative stress[1][3][4][6][7]. Emerging evidence suggests roles in disease processes such as neurodegeneration, ageing, and cancer, including a regulatory influence on epithelial-mesenchymal transition in cancer cells[3]. NUDT13 is part of a highly conserved enzyme family involved in cellular metabolism and nucleotide pool regulation[5]. No drugs, mechanisms of action for inhibitors, pharmacodynamic biomarkers, or specific safety concerns have been described for this target to date.
