SIRT5 is an NAD+-dependent protein lysine deacylase primarily localized in the mitochondrial matrix. It regulates various metabolic processes by removing succinyl, malonyl, and glutaryl groups from lysine residues on target proteins. It plays a role in glycolysis, the TCA cycle, fatty acid oxidation, the electron transport chain, ketone body formation, the urea cycle, nitrogenous waste management, and ROS detoxification. It is considered a potential drug target for treating metabolic disorders, cardiovascular diseases, cancer, and neurodegeneration.
