NADH:ubiquinone oxidoreductase subunit B1 (NDUFB1) is a small protein (approx. 7 kDa, 58 amino acids) encoded by the NDUFB1 gene on chromosome 14[1]. This protein is an accessory, non-catalytic subunit of mitochondrial complex I (NADH:ubiquinone oxidoreductase), a large enzyme complex in the mitochondrial inner membrane that is the first component of the electron transport chain[8][2]. NDUFB1 has a conserved L-shaped structure, anchoring complex I to the membrane through its hydrophobic domain and participating in interactions with other subunits through its hydrophilic domain[1]. While it is not directly involved in electron transfer catalysis, it contributes critically to the structural integrity, assembly, and function of complex I, ultimately supporting mitochondrial ATP production and energy metabolism[2][4][5]. Dysfunction or altered expression of NDUFB1 is associated with several diseases, especially those involving mitochondrial dysfunction, cancer, psychiatric, and metabolic disorders[1][2][5].
