NADH dehydrogenase (ubiquinone), commonly known as Complex I, is a multi-subunit enzyme complex found in the inner mitochondrial membrane and is the first and largest complex of the mitochondrial respiratory chain. It catalyzes the transfer of electrons from NADH to ubiquinone (Coenzyme Q), a process coupled with the translocation of protons across the mitochondrial membrane, thereby contributing to the proton gradient that drives ATP synthesis. The complex has an L-shaped structure, includes 40–50 subunits in animals and plants, and contains iron–sulfur centers and flavin mononucleotide (FMN) as cofactors. Dysfunction or inhibition of Complex I is implicated in numerous diseases, especially mitochondrial and neurodegenerative disorders, and is the target of several pharmacological and toxic agents[2][3][4][5][6][7][8].
