Overview

NADH dehydrogenase (ubiquinone) 1 beta subcomplex subunit 3 (NDUFB3) is an accessory subunit of mitochondrial respiratory chain Complex I, the largest of the five enzyme complexes in the electron transport chain. It is located in the inner mitochondrial membrane as a single-pass membrane protein, helping anchor Complex I and maintain its structural integrity. While it is not directly involved in electron catalysis, NDUFB3 is critical for complex assembly and function. It facilitates the initial transfer of electrons from NADH to coenzyme Q10, enabling ATP generation by oxidative phosphorylation. Mutations in NDUFB3 have clinical significance, being implicated in mitochondrial complex I deficiency and associated with disorders of oxidative phosphorylation. Altered NDUFB3 expression is also relevant in diseases such as cancer, where it can affect mitochondrial reactive oxygen species production and influence tumor progression and patient prognosis[1][2][4].
