Target intelligence / Profile preview
Natural killer T cell modulators are compounds or biologics that activate or inhibit the function of natural killer T (NKT) cells, a specialized subset of lymphocytes that bridge innate and adaptive immunity by responding to glycolipid antigens presented by CD1d molecules on antigen-presenting cells. Upon activation, NKT cells rapidly secrete large amounts of cytokines and chemokines, influencing downstream immune responses, tumor cell killing, and the balance between inflammation and tolerance. Therapeutic strategies leveraging NKT cell modulators include glycolipid agonists such as α-galactosylceramide and its analogs, experimental antibodies, cellular therapies, and vaccine vectors, all aimed at enhancing anti-tumor, anti-infective, or regulatory immune responses. The use of NKT cell modulators in therapy is complicated by risks of excessive inflammation, immune suppression, and loss of function following repeated stimulation.
Activation/inhibition of NKT cells via binding to CD1d-presented lipid antigens or modulation of the NKT cell surface receptors; Promotion of cytokine secretion and immune activation; Induction of NKT cell-dependent cytotoxicity of target (often tumor) cells; Immune tolerance or suppression via TH2 or regulatory cytokine skewing
