Necator americanus Glutathione-S-Transferase-1 (Na-GST-1) is a 24 kDa enzyme expressed by the human hookworm, a major cause of iron-deficiency anemia in tropical regions (Zhan et al., 2005, J Biol Chem). The enzyme is localized to the parasite's alimentary canal and plays a vital role in detoxifying host-derived heme, which is released during the digestion of blood (Asojo et al., 2007, J Mol Biol). By binding and sequestering these toxic heme molecules, Na-GST-1 prevents oxidative damage to the worm's internal tissues. Because this detoxification process is essential for the parasite's survival and fecundity, Na-GST-1 has been prioritized as a leading recombinant vaccine candidate (Bethony et al., 2011, Vaccine). Clinical development focuses on inducing host antibodies that neutralize the enzyme's catalytic and binding sites, effectively starving the parasite or causing lethal heme toxicity (Diemert et al., 2017, PLoS Negl Trop Dis). This target is particularly attractive because its structure is distinct from human glutathione S-transferases, minimizing the risk of autoimmune cross-reactivity.
The vaccine induces host IgG antibodies that bind to the Na-GST-1 enzyme in the hookworm gut, inhibiting its ability to detoxify heme and leading to parasite mortality or reduced fecundity (Diemert et al., 2017, PLoS Negl Trop Dis).
