Overview

CD112 (Nectin-2) and CD155 (Poliovirus receptor) are cell surface glycoproteins belonging to the nectin and nectin-like families of the immunoglobulin superfamily (UniProt P15151, Q92692). They function as ligands for a network of receptors on T cells and natural killer (NK) cells, including the inhibitory receptors TIGIT, PVRIG, and CD96, as well as the activating receptor CD226 (DNAM-1) (Frontiers in Immunology, 2022). In many cancers, CD112 and CD155 are overexpressed, leading to immune evasion by preferentially binding to inhibitory receptors and suppressing the cytotoxic activity of lymphocytes (NIH, 2024). Therapeutic strategies focus on blocking these inhibitory interactions using monoclonal antibodies like NTX1088 (anti-CD155) or by targeting their receptors with agents such as Tiragolumab (anti-TIGIT) and COM701 (anti-PVRIG) to restore anti-tumor immunity (AACR, 2023; Compugen). Additionally, CD155 serves as the entry receptor for poliovirus, a property exploited in the development of oncolytic viral therapies like PVSRIPO (MDPI, 2023).
