Negative regulator of antiviral response (NRAV) is a long non-coding RNA gene located on chromosome 12 (12q24.31), also referred to as DYNLL1-AS1 or DYNLL1 antisense RNA 1[7][6]. NRAV acts as a key negative modulator of the antiviral innate immune response, particularly by suppressing the transcription of multiple interferon-stimulated genes (ISGs) critical for defense against viral infection (e.g., IFITM3, MxA)[2]. During infection with various RNA and DNA viruses, NRAV levels are rapidly downregulated as part of the host's antiviral defense, and its ectopic overexpression promotes viral replication and impairs defense[2][5]. Mechanistically, NRAV influences antiviral gene expression by affecting histone modification at ISG promoters. While NRAV is not a receptor, enzyme, or traditional 'druggable' protein, it is viewed as a potential therapeutic target due to its prominent modulatory role in host–virus interactions and immune regulation[2][3][5].
