Overview

The polysaccharide capsule of *Neisseria meningitidis* is a crucial virulence factor that enables the bacterium to evade host immune responses and cause invasive diseases. Vaccines targeting the capsular polysaccharides of serogroups A, C, Y, and W-135 are highly effective in preventing meningococcal disease caused by these serotypes. The capsule consists of unique repeating polysaccharide units specific to each serogroup, which determine immunogenicity. These capsules are anchored to the outer membrane via lipid modification and diacylglycerols at their reducing ends. Quadrivalent vaccines targeting these four serogroups use purified capsular polysaccharides from each group as antigens.
