Neisseria meningitidis Porin A (PorA) is a major integral outer membrane protein that functions as a selective channel for the transport of nutrients and ions across the bacterial cell wall (UniProt P0A316). It is highly immunogenic and serves as a primary target for bactericidal antibodies, making it a critical component in the development of vaccines against serogroup B meningococci, which lack a suitable polysaccharide target (PubMed 15845118). The P1.4 designation refers to a specific variable region (VR2) of the PorA protein that was the dominant antigen in the MeNZB vaccine used to combat a specific epidemic strain in New Zealand (PubMed 16309314). Outer Membrane Vesicles (OMVs) are naturally occurring blebs from the bacterial surface that contain PorA along with other antigens like PorB and various lipoproteins, providing a complex antigenic profile that mimics the native bacterial surface (PubMed 22244487). In modern multicomponent vaccines such as 4CMenB (Bexsero), OMVs from the NZ98/254 strain expressing PorA P1.4 are utilized to enhance the breadth of the protective immune response against diverse meningococcal strains (FDA Bexsero Label).
