Overview

Neugrin, neurite outgrowth associated (NGRN) is a mitochondrial-associated protein involved in the biogenesis and assembly of mitochondrial ribosomes[1][3][4][5][6]. It acts as a component of a multi-protein/RNA module—alongside proteins such as RCC1L, RPUSD3, RPUSD4, TRUB2, FASTKD2, and 16S mitochondrial ribosomal RNA—regulating the abundance of 16S mt-rRNA and required for intra-mitochondrial translation of core subunits essential for mitochondrial oxidative phosphorylation[1][3][4][5]. NGRN displays rRNA binding activity, positively influences mitochondrial translation, and is localized to the mitochondrion, mitotic spindle, and nuclear body[4][7]. Human diseases linked to NGRN include a subset of hereditary deafness, but only limited association with broader pathophysiological processes has been reported to date[4]. NGRN is not a receptor, enzyme, transporter, or recognized direct therapeutic target, and currently there are no known drugs or clinical biomarker assays targeting this protein[4][6].
