Neuronal potassium channels are a diverse superfamily of transmembrane proteins that selectively facilitate the efflux of potassium ions across the neuronal cell membrane. They are fundamental to the nervous system, where they maintain the resting membrane potential, modulate the duration and frequency of action potentials, and regulate neurotransmitter release (StatPearls, 2023). These channels are classified into several families, including voltage-gated (Kv), calcium-activated (KCa), inwardly rectifying (Kir), and two-pore domain (K2P) channels (PubMed, 2021). Dysfunction in these channels, often due to genetic mutations or autoimmune responses, is associated with a wide range of channelopathies such as epilepsy, episodic ataxia, and neuropathic pain (NIH, 2022). Consequently, they are major therapeutic targets; for instance, KCNQ (Kv7) channel openers like retigabine are used as anticonvulsants, while blockers like 4-aminopyridine are used to improve walking in multiple sclerosis patients (PubChem, 2024).
