Overview

Neuropilin-1 (NRP1) receptor pre-messenger RNA is the primary transcript of the NRP1 gene, which encodes a critical transmembrane co-receptor involved in various signaling pathways. The resulting NRP1 protein acts as a co-receptor for vascular endothelial growth factor (VEGF) and class 3 semaphorins, playing a pivotal role in angiogenesis, axonal guidance, and immune cell regulation [UniProt: P35052]. In pathological states, particularly in oncology, NRP1 is often overexpressed, promoting tumor vascularization, cell survival, and metastasis [NCBI Gene: 8829]. Furthermore, NRP1 has been identified as a significant co-factor for SARS-CoV-2 entry into host cells, enhancing viral infectivity [PMID: 32511329]. Targeting the pre-mRNA with antisense oligonucleotides (ASOs) or splice-switching molecules allows for the modulation of protein levels or the induction of soluble isoforms (sNRP1) that act as decoys, providing a therapeutic strategy to inhibit tumor growth and viral infection [PMID: 25650324, PMID: 17210688]. This approach offers a high degree of specificity compared to traditional small molecules or antibodies by directly intervening at the genetic processing level.
