NY-ESO-1–derived peptide–HLA complexes are specialized molecular assemblies consisting of fragments from the New York esophageal squamous cell carcinoma 1 (NY-ESO-1) protein bound to Human Leukocyte Antigen (HLA) molecules. These complexes are presented on the surface of dendritic cells (DCs) to initiate and modulate the adaptive immune response against cancer cells (PubMed: 31515463). NY-ESO-1 is a well-characterized cancer-testis antigen that is typically expressed in germ cells but aberrantly upregulated in various cancers, including synovial sarcoma and melanoma (NCI, 2024). When DCs internalize NY-ESO-1, they process it into peptides and present them via HLA Class I or II molecules to T-cell receptors (TCRs) on CD8+ and CD4+ T cells, respectively. This interaction is a critical step in the cancer-immunity cycle, leading to the expansion of tumor-specific cytotoxic T lymphocytes. Therapeutic interventions targeting these complexes include TCR-engineered T-cell therapies, such as afamitresgene autoleucel (Tecelra), which was recently FDA-approved for synovial sarcoma (FDA, 2024). Additionally, dendritic cell-based vaccines like LV305 are designed to enhance the presentation of these specific antigens to boost endogenous immunity. Because NY-ESO-1 expression is restricted to immune-privileged sites in healthy individuals, these complexes represent highly specific targets for precision oncology.
