Non-small cell lung cancer (NSCLC) tumor-associated antigens (TAAs) are a heterogeneous group of molecules, including proteins, glycoproteins, and glycolipids, that are either uniquely expressed or significantly overexpressed in lung cancer cells compared to healthy tissues (PubMed: PMC6343131). These antigens, such as MUC1, CEA, and NY-ESO-1, are involved in various biological processes including cell adhesion, signal transduction, and immune evasion (UniProt). In clinical oncology, TAAs are utilized as targets for immunotherapies like therapeutic vaccines (e.g., TG4010, CIMAvax-EGF) and monoclonal antibodies designed to stimulate the host's immune system to recognize and destroy malignant cells (NIH). The identification and targeting of these antigens are central to the development of personalized medicine in NSCLC, allowing for more precise therapeutic interventions. However, challenges such as antigen heterogeneity within the tumor and the risk of on-target off-tumor toxicity remain significant hurdles in the development of effective TAA-targeted therapies (PubMed: PMC7464943). By targeting these antigens, researchers hope to overcome the immunosuppressive environment of the lung tumor microenvironment.
