Overview

Non-variola orthopoxvirus DNA refers to the double-stranded DNA genome of viruses within the Orthopoxvirus genus, excluding the Variola virus which causes smallpox (CDC, 2022). This group includes several zoonotic pathogens of significant public health concern, most notably the Monkeypox virus (Mpox), as well as the Cowpox virus and Vaccinia virus (NIH, 2023). The DNA genome serves as the primary repository of genetic information, acting as the essential template for both transcription of viral mRNA and replication of the viral genome within the host cell's cytoplasm (Siegrist & Johnston, 2009). In clinical diagnostics, this DNA is the specific molecular target for real-time PCR assays, such as the CDC Non-variola Orthopoxvirus (NVO) test, which is used to identify infections during outbreaks (CDC, 2022). While most antiviral drugs target viral proteins, the replication of this DNA is the ultimate process inhibited by therapeutics like Cidofovir and Brincidofovir (FDA, 2021). These drugs act as nucleotide analogs that are incorporated into the nascent DNA strand by the viral DNA polymerase, leading to premature chain termination and the cessation of viral production (PubChem, 2024). Monitoring the presence and quantity of this DNA in clinical samples is a critical biomarker for diagnosing infections and assessing the efficacy of antiviral treatments (Lanford et al., 2022).
