The norepinephrine transporter is a sodium-chloride dependent membrane protein (encoded by SLC6A2), restricted to noradrenergic neurons, that clears norepinephrine from the synaptic cleft via reuptake, regulating neurotransmission, mood, and attention[1][4]. The serotonin transporter (SLC6A4) performs analogous reuptake function for serotonin[2][3]. Both are major drug targets for antidepressants (SNRIs, tricyclics) and are implicated in a range of psychiatric and neurological conditions[2][6]. Drug binding sites in these transporters overlap with substrate binding pockets, and genetic or drug-induced modifications can alter their clinical efficacy and safety profiles[2][3][4].
Inhibition of reuptake of norepinephrine and/or serotonin, increasing their extracellular concentrations in the synaptic cleft. Certain drugs selectively inhibit one or both transporters, altering neurotransmission involved in mood and pain regulation.
