Overview

Norovirus capsid protein VP1 is the major structural protein forming the outer shell of noroviruses, including the GI.1 genotype (Norwalk virus). It plays a central role in viral assembly, host cell attachment, immune recognition, and genetic diversity. VP1 self-assembles into an icosahedral capsid and is comprised of Shell (S) and Protruding (P) domains. The P domain is responsible for binding to histo-blood group antigens (HBGAs) on host cells. Variability in the P2 subdomain contributes to immune evasion and strain diversity. VP1 interacts with minor capsid protein VP2 to assist genome encapsidation and regulates viral genome synthesis. It is a target for neutralizing antibodies and vaccine development.
