Overview

The Norovirus Genogroup II genotype 3 (GII.3) Major Capsid Protein (VP1) is the primary structural component of the virus, responsible for forming the icosahedral shell that protects the viral RNA. VP1 is divided into two main domains: the conserved Shell (S) domain, which forms the structural core, and the Protruding (P) domain, which extends outward and mediates interaction with host cell receptors, specifically Histo-blood group antigens (HBGAs). GII.3 is a major cause of endemic acute gastroenteritis, particularly in children, and exhibits significant genetic diversity and periodic antigenic shifts. As the principal surface-exposed protein, VP1 is the primary target for the development of vaccines, such as virus-like particles (VLPs), and neutralizing monoclonal antibodies intended to block viral entry into intestinal epithelial cells. There are currently no FDA-approved antivirals or vaccines specifically for GII.3, but it remains a critical target in multivalent vaccine candidates and entry-inhibitor research.
