Immune checkpoint modulators are molecules expressed on T cells that regulate immune responses. These checkpoints can either stimulate or inhibit T cell activity, playing a crucial role in maintaining self-tolerance and modulating the intensity and duration of immune responses. Tumors often exploit inhibitory checkpoint pathways to evade immune surveillance by suppressing T cell activation. Targeting these molecules with monoclonal antibodies or other agents can "release the brakes" on anti-tumor immunity. Novel modulators are being explored for potential in combination therapies and minimizing autoimmune side effects while maximizing anti-tumor efficacy.
