Overview

The NR2B subunit-containing NMDA receptor is a subtype of the ionotropic glutamate receptor, specifically a variant that incorporates the NR2B (GluN2B or GRIN2B) subunit. It plays a critical role in synaptic transmission, plasticity, and various physiological and pathological processes in the central nervous system. It is a heterotetramer composed of two GluN1 subunits and two NR2B subunits, forming a ligand-gated ion channel permeable to Ca²⁺, Na⁺, and K⁺. It is implicated in learning, memory, pain modulation, and circadian rhythm, and is linked to neurological disorders like chronic pain, neurodegenerative diseases, and psychiatric conditions. Selective antagonists targeting this subunit have shown promise for therapeutic intervention.
