Overview

Nuclear antigens are a heterogeneous group of molecules, including proteins, DNA, and RNA, that are localized within the cell nucleus and are essential for genetic maintenance and expression. In clinical medicine, these antigens are primarily significant as the targets of autoantibodies in systemic autoimmune rheumatic diseases (SARDs). The presence of antinuclear antibodies (ANAs) directed against these components is a key diagnostic marker for conditions like systemic lupus erythematosus (SLE), Sjögren's syndrome, and systemic sclerosis (NIH, MedlinePlus). While nuclear antigens are not direct therapeutic targets for small molecules or biologics, the immune pathways that react to them are the focus of various immunomodulatory treatments. Therefore, they are primarily utilized as biomarkers for diagnosis and disease classification rather than as druggable receptors or enzymes.
