Overview

NF-κB p65 mRNA is the messenger RNA transcript of the RELA gene, which encodes the p65 (RelA) subunit of the Nuclear Factor kappa-B (NF-κB) transcription factor complex (UniProt P19838). As a central component of the canonical NF-κB pathway, the p65 protein regulates the transcription of a vast array of genes involved in the innate and adaptive immune responses, cell survival, and inflammation (PubMed: 24336188). In various diseases, particularly chronic inflammatory conditions and cancers, the NF-κB pathway is pathologically overactivated, leading to the sustained production of pro-inflammatory cytokines and resistance to apoptosis (PubMed: 12486457). Targeting the p65 mRNA directly using RNA interference (RNAi) or antisense oligonucleotides (ASOs) allows for the specific knockdown of p65 protein synthesis, offering a therapeutic strategy to suppress aberrant NF-κB signaling (PubMed: 15123770). This approach is being explored to treat conditions such as rheumatoid arthritis, inflammatory bowel disease, and solid tumors. However, challenges remain regarding the efficient delivery of RNA-based therapeutics to target tissues and the potential for systemic toxicity or unintended immunosuppression due to the essential role of NF-κB in normal cellular homeostasis (PubMed: 16751388).
