Overview

Nuclear poly(ADP-ribose) polymerase 1 (PARP-1) is a multifunctional nuclear enzyme that plays critical roles in DNA repair, chromatin remodeling, transcription regulation, and cell death pathways. It detects DNA strand breaks and synthesizes poly(ADP-ribose) (PAR) chains, signaling DNA damage and recruiting repair proteins. PARP-1 is involved in multiple DNA repair pathways and regulates transcription/chromatin structure. Inhibition of PARP-1 is a therapeutic strategy, particularly in cancers with BRCA mutations or other homologous recombination deficiencies.
