Overview

Nuclear protein 1, also known as NUPR1, is a small, highly conserved chromatin-binding transcription factor that is rapidly induced by various forms of cellular stress. It plays a critical role in converting stress signals into specific gene expression programs that enhance cell survival under adverse conditions. In the context of pancreatic cancer and other malignancies, NUPR1 is frequently upregulated and contributes to tumor development by promoting resistance to apoptosis and supporting adaptation to nutrient deprivation or other microenvironmental stresses. Loss or inhibition of NUPR1 sensitizes cells to apoptotic stimuli and impairs their ability to survive under metabolic or genotoxic stress. Beyond its role in cancer biology, NUPR1 has been implicated in processes such as autophagy regulation and DNA repair following double-strand breaks. Its activity links it with several disease states where cellular adaptation to stress is crucial for pathogenesis or progression.
