Overview

The Nuclear ubiquitous casein and cyclin-dependent kinase substrate 1 (NUCKS1) mRNA 3' untranslated region (3' UTR) is a critical regulatory element that governs the expression of the NUCKS1 protein, a key player in DNA repair and cell cycle regulation (UniProt: Q9H1E3). This 3' UTR contains multiple binding sites for microRNAs, such as miR-137, which normally suppress NUCKS1 expression; however, the loss of this regulation is frequently associated with the progression of various cancers, including breast and colorectal cancer (PubMed: 30217975, PubMed: 25333241). Beyond oncology, the NUCKS1 transcript is implicated in metabolic pathways, where its levels influence insulin signaling and obesity (PubMed: 25605870). From a therapeutic perspective, the NUCKS1 mRNA 3' UTR is a target for RNA-based interventions, such as antisense oligonucleotides (ASOs) or microRNA mimics, designed to downregulate the protein in disease states. While no small-molecule drugs currently target this region, it represents a significant opportunity for precision oncology and metabolic disease management. Challenges for this target include ensuring tissue-specific delivery and minimizing off-target hybridization. Overall, the NUCKS1 mRNA 3' UTR is a vital node in the post-transcriptional landscape of cancer and metabolic disease.
