NLRX1 (Nucleotide-binding oligomerization domain, leucine rich repeat containing X1) is an intracellular NOD-like pattern recognition receptor distinct from other inflammasome-associated NLR proteins. It inhibits pro-inflammatory pathways, including NF-κB and type-I interferon signaling, and is involved in negative regulation of innate immune responses, ROS production, autophagy, and cell death. NLRX1 functions as a key modulator of inflammation and immune signaling through scaffolding of multiprotein complexes but does not form canonical inflammasomes. Altered expression or function of NLRX1 is linked to susceptibility and severity of various inflammatory and infectious diseases, making it an emerging target for immunomodulatory drug discovery[1][2][3][6].
